the defence is removed from the base defence of the enemys. Ie. if you look through monstats.txt, the values for defence are rather low (200-400 average iirc)
That base number is then multiplied by a value in monlvl.txt.
Say the multiplier is 1500% and the enemy has 500 defence in monstats.txt. The enemy will have 8000 defence in game.
The flat -275 defence is removed from the 500 base the enemy has. So instead of 1500% of 500 defence, its 1500% of 225 defence (3600 defence)
-xxx enemy defence per hit works in the same way too as far as i know
